Collection:

Collection ID:CO002244
Collection Summary:Animals were acclimatized for 48 hrs in polypropylene cages. Urine samples were collected at 24 hrs post radiation exposure.
Sample Type:Urine
Storage Conditions:-80℃
Collection Tube Temp:4℃

Treatment:

Treatment ID:TR002263
Treatment Summary:Random categorization was done into three groups: control, 5 Gy (sub-lethal) and 7.5 Gy (lethal) irradiated (seven animals in each group). Mice were kept for 12 hrs light and dark cycle with room temperature (19–23°C) and humidity (45–65%) conditions properly maintained. Mice were exposed to total-body gamma radiation (field size; 35x35 cm) of 5 Gy and 7.5 Gy through Tele 60Co unit γ- irradiation facility (Bhabhatron II, Panacea Medical, Bangalore, India) with dose rate of 1 Gy/min. Control mice were sham irradiated.
Treatment:Radiation exposure
Treatment Dose:5 Gy and 7.5 Gy Gamma-60Cobalt irradiation

Sample Preparation:

Sampleprep ID:SP002257
Sampleprep Summary:Urine samples (50 uL) were added to 200ul of water containing internal standards (caffeine, nitrobenzoic acid and acetaminophen) and were filtered through 3 kDa filter.
Processing Storage Conditions:On ice
Extract Storage:On ice

MS:

MS ID:MS003305
Analysis ID:AN003547
Instrument Name:ABI Sciex 5600 TripleTOF
Instrument Type:Triple TOF
MS Type:ESI
MS Comments:Samples were ionized using Electron Spray Ionization(ESI) in both the modes. The various MS parameters used for the present study are as discussed. The gas flow, GS1 and GS2 were kept at 50 psi, curtain gas flow (CUR) at 30 psi and capillary voltage(ISVF) at 5500 V for positive and - 4500 V for negative ionization mode with source temperature 400 0C for both ionization modes. The MS calibration was performed through CDS (calibrant delivery system). For MS/MS fragmentation of metabolites, collision energy was set at 35 eV with a collision energy spread of ± 15 eV and declustering potential of 60 eV. Mass range was set at 100- 1200m/z. Metabolites were identified by matching with, company provided library and Metlin public database (https://metlin.scripps.edu).
