PR:PROJECT_SUMMARY               	Colorectal cancer (CRC) is one of the major leading cause of death in humans
PR:PROJECT_SUMMARY               	worldwide. In Malaysia, CRC is the fourth leading cause of death, covered 13.2%
PR:PROJECT_SUMMARY               	of new cancer diagnoses in Peninsular Malaysia in 2006. Causable factors are
PR:PROJECT_SUMMARY               	remaining unknown. Changes in dietary may alter colonic microbiome and mucosal
PR:PROJECT_SUMMARY               	immune microenvironment inducing colon oncogenesis. Previous investigation
PR:PROJECT_SUMMARY               	focused on mutagen production by colonic bacteria or their conversion of dietary
PR:PROJECT_SUMMARY               	procarcinogens into DNA-damaging molecules. Yet, no direct links between the
PR:PROJECT_SUMMARY               	metabolic activities of bacteria and sporadic CRC were identified. The objective
PR:PROJECT_SUMMARY               	of this study was to prospectively characterize the human colonic microbiome in

ST:STUDY_TITLE                   	Metabolomics and 16S rRNA sequencing of human colorectal cancers and adjacent

ST:STUDY_SUMMARY                 	Colorectal cancer (CRC) is ranked the third most common cancer in human
ST:STUDY_SUMMARY                 	worldwide. However, the exact mechanisms of CRC are not well established.
ST:STUDY_SUMMARY                 	Furthermore, there may be differences between mechanisms of CRC in the Asian and
ST:STUDY_SUMMARY                 	in the Western populations. In the present study, we utilized a liquid
ST:STUDY_SUMMARY                 	chromatography-mass spectrometry (LC-MS) metabolomic approach supported by the
ST:STUDY_SUMMARY                 	16S rRNA next-generation sequencing to investigate the functional and
ST:STUDY_SUMMARY                 	taxonomical differences between paired tumor and unaffected (normal) surgical
ST:STUDY_SUMMARY                 	biopsy tissues from 17 Malaysian patients. Metabolomic differences associated
ST:STUDY_SUMMARY                 	with steroid biosynthesis, terpenoid biosynthesis and bile metabolism could be
ST:STUDY_SUMMARY                 	attributed to microbiome differences between normal and tumor sites. The
ST:STUDY_SUMMARY                 	relative abundances of Anaerotruncus, Intestinimonas and Oscillibacter displayed
ST:STUDY_SUMMARY                 	significant relationships with both steroid biosynthesis and terpenoid and
ST:STUDY_SUMMARY                 	triterpenoid biosynthesis pathways. Metabolites involved in serotonergic
ST:STUDY_SUMMARY                 	synapse/ tryptophan metabolism (Serotonin and 5-Hydroxy-3-indoleacetic acid
ST:STUDY_SUMMARY                 	[5-HIAA]) were only detected in normal tissue samples. On the other hand,
ST:STUDY_SUMMARY                 	S-Adenosyl-L-homocysteine (SAH), a metabolite involves in methionine metabolism
ST:STUDY_SUMMARY                 	and methylation, was frequently increased in tumor relative to normal tissues.
ST:STUDY_SUMMARY                 	In conclusion, this study suggests that local microbiome dysbiosis may
ST:STUDY_SUMMARY                 	contribute to functional changes at the cancer sites. Results from the current
ST:STUDY_SUMMARY                 	study also contributed to the list of metabolites that are found to differ
ST:STUDY_SUMMARY                 	between normal and tumor sites in CRC and supported our quest for understanding
ST:STUDY_SUMMARY                 	the mechanisms of carcinogenesis.
