PR:PROJECT_SUMMARY               	Alzheimer’s disease (AD) is a devastating neurodegenerative disorder that robs
PR:PROJECT_SUMMARY               	people of their memory and cognitive function. Currently, no successful
PR:PROJECT_SUMMARY               	treatment or preventative measure exists for AD. Calorie restriction (CR) is a
PR:PROJECT_SUMMARY               	dietary regimen posited to suppress genetic programs of aging and reduce
PR:PROJECT_SUMMARY               	AD-related pathology. CR is known to enhance longevity and mitigate aging
PR:PROJECT_SUMMARY               	phenotypes in multiple model species. Mechanisms underlying the benefits of CR
PR:PROJECT_SUMMARY               	remain unknown, particularly in areas of the brain selectively vulnerable to
PR:PROJECT_SUMMARY               	age-related AD pathology such as the hippocampus, a region crucial for learning
PR:PROJECT_SUMMARY               	and memory. Moreover, AD pathology can be influenced by changes in diet,
PR:PROJECT_SUMMARY               	metabolism, and immunity, indicating that factors distant from the brain may
PR:PROJECT_SUMMARY               	play a role in pathogenesis. The intestinal microbiota, composed of trillions of
PR:PROJECT_SUMMARY               	microbial cells, influences host metabolism, immunity, and cognitive function,
PR:PROJECT_SUMMARY               	and is posited to be linked mechanistically to AD pathobiology, but a specific
PR:PROJECT_SUMMARY               	role remains to be adequately tested. We hypothesize that mechanisms underlying
PR:PROJECT_SUMMARY               	the benefits of CR are cell-type and organ specific, involving the gut-brain
PR:PROJECT_SUMMARY               	microbiome throughout the lifespan, this requiring subregional analysis in the
PR:PROJECT_SUMMARY               	brain as well as coordinated assessments of key peripheral targets including the
PR:PROJECT_SUMMARY               	liver, fecal pellets , and plasma. Thus, CR is proposed to be a viable treatment
PR:PROJECT_SUMMARY               	option that may ameliorate the development of AD-related pathology, and
PR:PROJECT_SUMMARY               	importantly, reveal mechanisms that attenuate age-related expression changes in
PR:PROJECT_SUMMARY               	vulnerable cells.

ST:STUDY_SUMMARY                 	This pilot metabolomic study will evaluate brain specimens from an established
ST:STUDY_SUMMARY                 	mouse model of AD, the tq2576 mouse model of cerebral amyloid overexpression
ST:STUDY_SUMMARY                 	(APP), in comparison to their non-transgenic (NTG) littermates. These animals
ST:STUDY_SUMMARY                 	were either on a CR or ad libitum (AL) diet, and specimens were collected at two
ST:STUDY_SUMMARY                 	time points (5 and 15 months of age). Tissue from this cohorts of mice have
ST:STUDY_SUMMARY                 	already undergone microbiome analysis, and await coordinated brain and
ST:STUDY_SUMMARY                 	peripheral tissue assessments. Future analysis will include metabolomics,
ST:STUDY_SUMMARY                 	RNA-seq, and microarray data to assess the gut-brain microbiome system in

SP:SAMPLEPREP_SUMMARY            	Brain tissue samples were homogenized in 50:50 Acetonitrile:Water , and the
SP:SAMPLEPREP_SUMMARY            	final concentration of all of the extracts was 1 mL/mg. Of the 71 samples, 48
SP:SAMPLEPREP_SUMMARY            	had sufficient volume for study samples and for an analytical quality control
SP:SAMPLEPREP_SUMMARY            	(QC) total pool sample. This total pool was generated by transferring a 50 µL
SP:SAMPLEPREP_SUMMARY            	of each sample and aliquoting into six replicates, which were processed
SP:SAMPLEPREP_SUMMARY            	identically to the study samples. A 250 µL aliquot of each sample homogenate
SP:SAMPLEPREP_SUMMARY            	was lyophilized and then reconstituted with 250 µL of Phosphate buffer with
SP:SAMPLEPREP_SUMMARY            	Chenomx ISTD and 6mM Imidazole. Samples were vortexed and centrifuged before
SP:SAMPLEPREP_SUMMARY            	transferring 225 µL of each sample into pre-labeled 3mm NMR tubes for data
SP:SAMPLEPREP_SUMMARY            	acquisition on a 700 MHz spectrometer.
