PR:PROJECT_SUMMARY               	example urine) can provide a profile of the metabolic state of an organism.
PR:PROJECT_SUMMARY               	Metabolomics is a powerful technique for revealing the impact of exposure on the
PR:PROJECT_SUMMARY               	overall biochemistry of an individual or system. Opioids can modify the output
PR:PROJECT_SUMMARY               	of urinary metabolites through many integrated neural and hormonal mechanisms
PR:PROJECT_SUMMARY               	within the periphery, central nervous system, and kidneys. Opioids modulate the
PR:PROJECT_SUMMARY               	expression of genes involved in neuroplasticity through epigenetic and possibly
PR:PROJECT_SUMMARY               	RNA modifications, ultimately change the intracellular signaling cascades and
PR:PROJECT_SUMMARY               	dysfunction, and cause long-lasting changes in metabolome. The objective of this
PR:PROJECT_SUMMARY               	study is to identify how opium impacts metabolic pathways to provide markers of
PR:PROJECT_SUMMARY               	abuse, long-term opium addiction, the addiction molecular pathway, and unknown
PR:PROJECT_SUMMARY               	metabolites that are important to differentiation of the study phenotypes. To
PR:PROJECT_SUMMARY               	reach these goals in the present study, the urine specimens of opium abusers and
PR:PROJECT_SUMMARY               	non-users as controls will be profiled using an untargeted liquid chromatography
PR:PROJECT_SUMMARY               	mass spectrometric (LC-MS/MS) at University of North Carolina at Chapel Hill.
PR:PROJECT_SUMMARY               	The Golestan Cohort Study is conducted in Northeast of Iran to primarily study
PR:PROJECT_SUMMARY               	the risk factors for upper gastrointestinal cancers in this high-risk region, in
PR:PROJECT_SUMMARY               	which about 50,000 volunteers were analyzed for opium users and their mortality.
PR:PROJECT_SUMMARY               	More than 8,000 of participants (17%) age 40-75 reported opium use with a mean
PR:PROJECT_SUMMARY               	duration of 12.7 years. Opium was either smoked or orally consumed. The
PR:PROJECT_SUMMARY               	participants were selected from the cohort stratified by opium use patterns and
PR:PROJECT_SUMMARY               	tobacco use.

SP:SAMPLEPREP_SUMMARY            	Urine samples were allowed to thaw at 4 °C overnight. All samples were vortexed
SP:SAMPLEPREP_SUMMARY            	via multiple tube vortex for 2 min at 3000 rpm. The urine were spin downed using
SP:SAMPLEPREP_SUMMARY            	centrifuge at 1000 rpm for 30 sec. A volume of 7 µL aliquot of all 298 samples
SP:SAMPLEPREP_SUMMARY            	were transferred into one 10.0 mL tube labeled as pooled. A volume of 50 µL of
SP:SAMPLEPREP_SUMMARY            	urine (including study pool) was transferred to a new, pre-labeled Lo-Bind
SP:SAMPLEPREP_SUMMARY            	Eppendorf tube. A volume of 400 µL Protein Precipitation Buffer with Internal
SP:SAMPLEPREP_SUMMARY            	Standard (500 ng/ml Tryptophan-d5 in methanol) was added to each tube. All
SP:SAMPLEPREP_SUMMARY            	individual samples were thoroughly mixed on multiple tube vortexer for 2 mins at
SP:SAMPLEPREP_SUMMARY            	5000 rpm. The protein precipitate was palleted using a centrifuge operating at
SP:SAMPLEPREP_SUMMARY            	room temperature and at 16,000 rcf for 4 min. A volume of 350 µl of the
SP:SAMPLEPREP_SUMMARY            	supernatant was transferred into pre-labeled 2.0 ml Low-bind Eppendorf tube.
SP:SAMPLEPREP_SUMMARY            	Sample were dried using speed vac overnight. A volume of 100 µL of
SP:SAMPLEPREP_SUMMARY            	reconstitution buffer (95:5 Water-Methanol) was added to each sample. Samples
SP:SAMPLEPREP_SUMMARY            	were thoroughly mixed on multiple tube vortexer for 10 mins at 5000 rpm. Samples
SP:SAMPLEPREP_SUMMARY            	were centrifuged at 4°C and at 16,000 rcf for 10 min. The supernatant was
SP:SAMPLEPREP_SUMMARY            	transferred to pre-labeled autosampler vials. A volume of 5 µL was injected
SP:SAMPLEPREP_SUMMARY            	onto column.
