PR:PROJECT_TITLE                 	Reducing the metabolic burden of rRNA synthesis promotes healthy longevity
PR:PROJECT_SUMMARY               	Ribosome biogenesis is an anabolic process driven by RNA polymerase I (Pol
PR:PROJECT_SUMMARY               	I)-mediated synthesis of pre-ribosomal RNA (pre-rRNA). While Pol I activity was
PR:PROJECT_SUMMARY               	previously linked to longevity, the underlying mechanisms were not studied
PR:PROJECT_SUMMARY               	beyond effects on protein translation and downstream proteostasis. Here we used
PR:PROJECT_SUMMARY               	multi-omics and functional tests to show that curtailment of Pol I activity
PR:PROJECT_SUMMARY               	preserves mitochondrial function and lowers ATP expenditure, thereby promoting
PR:PROJECT_SUMMARY               	longevity in Caenorhabditis elegans. Reduced pre-rRNA synthesis also improved
PR:PROJECT_SUMMARY               	longevity and energy homeostasis in Drosophila melanogaster and human cells,
PR:PROJECT_SUMMARY               	respectively. Conversely, the enhancement of pre-rRNA synthesis boosted growth
PR:PROJECT_SUMMARY               	and neuromuscular performance of young nematodes at the cost of accelerated
PR:PROJECT_SUMMARY               	metabolic decline, mitochondrial stress and premature aging. Moreover,
PR:PROJECT_SUMMARY               	restriction of Pol I activity extended lifespan more potently than translational
PR:PROJECT_SUMMARY               	repression, and retained its geroprotective effects when initiated late in life,
PR:PROJECT_SUMMARY               	showcasing moderation of Pol I activity as an effective longevity treatment not
PR:PROJECT_SUMMARY               	limited by aging.
