While two genes (β-catenin and adenomatous polyposis coli) PR:PROJECT_SUMMARY have been found in patients who develop desmoids, it is unclear how these PR:PROJECT_SUMMARY mutations and other downstream mechanisms lead to desmoid tumorigenesis. PR:PROJECT_SUMMARY Extensive research has been explored in the molecular biology of desmoids; PR:PROJECT_SUMMARY however, the use of metabolomics to understand the how the low molecular weight PR:PROJECT_SUMMARY complements of cells, tissues, and biological fluids are perturbed by this PR:PROJECT_SUMMARY highly localized disease. Additionally, the Desmoid Collaboration for a Cure has PR:PROJECT_SUMMARY identified 45 active drugs against primary cell lines. Monolayer cell cultures were grown in DMEM supplemented with 5% TR:TREATMENT_SUMMARY fetal bovine serum and maintained at 37°C in 5% CO2. Cells were divided when TR:TREATMENT_SUMMARY confluent and experiments were performed between the third and sixth passages. TR:TREATMENT_SUMMARY Approximately 10 x 106 cells were treated with 1.0uM Dasatinib (Selleck, TR:TREATMENT_SUMMARY Houston, USA) dissolved in DMSO, or 0.5uM FAK Inhibitor 14 (Cayman Chemicals TR:TREATMENT_SUMMARY Company, Michigan, USA) dissolved in water. Cells were incubated in fresh media TR:TREATMENT_SUMMARY containing the inhibitors, or vehicle, for 24 hours. #SAMPLEPREP SP:SAMPLEPREP_SUMMARY Cell media was extracted with 50% acetonitrile:water, vortexed, and centrifuged.
