PM and MetSyn are strong risk factors for PR:PROJECT_SUMMARY chronic obstructive pulmonary disease (COPD) and asthma. After the World Trade PR:PROJECT_SUMMARY Center collapse in 9/11/2001, PM-exposed individuals from the Fire Department of PR:PROJECT_SUMMARY New York City (FDNY) developed a progressively lung disease. This nested PR:PROJECT_SUMMARY case-cohort study is composed of never smoking, WTC exposed firefighters with PR:PROJECT_SUMMARY normal pre-9/11 lung function presenting for subspecialty pulmonary evaluation PR:PROJECT_SUMMARY (SPE) before March 2008. Representative cohort controls with serum drawn within PR:PROJECT_SUMMARY six months of 9/11 (n=100). FEV1 at subspecialty exam defined cases: susceptible PR:PROJECT_SUMMARY World Trade Center Lung Injury (WTC-LI) cases (n=50) had FEV1< lower limit of PR:PROJECT_SUMMARY normal (LLN) and resistant WTC-LI cases with FEV1 ≥107% predicted (n=50). This PR:PROJECT_SUMMARY study will determine the metabolomics profile that differentiates firefighters PR:PROJECT_SUMMARY with WTC-LI, firefighters resistant to WTC-LI, and similarly exposed cohort PR:PROJECT_SUMMARY controls. Serum processing and analysis occurred in the PI’s Laboratory SP:SAMPLEPREP_SUMMARY at New York University /Bellevue. The specimens were stored in the New York SP:SAMPLEPREP_SUMMARY University-The Fire Department of New York City- World Trade Center Lung Injury SP:SAMPLEPREP_SUMMARY Biorepository (NYU-FDNY WTC) in Dr. Nolan’s laboratory. Each stored specimen SP:SAMPLEPREP_SUMMARY was labeled with a unique code. This unique code on the sample was linked to a SP:SAMPLEPREP_SUMMARY specific subject. Samples were stored in -80 oC and shipped to the Eastern SP:SAMPLEPREP_SUMMARY Regional Comprehensive Metabolomics Resource Core (ERCMRC) on dry ice. Table 1 SP:SAMPLEPREP_SUMMARY lists the sample ID and associated metadata provided. Serum samples were thawed SP:SAMPLEPREP_SUMMARY at 4 °C overnight. All samples were vortexed via multiple tube vortex mixer for SP:SAMPLEPREP_SUMMARY 2 min at 3000 rpm. Serum samples were centrifuged at 1000 rpm for 30 sec. A SP:SAMPLEPREP_SUMMARY volume of 8 µL aliquot of each studied samples was transferred into a 10.0 mL SP:SAMPLEPREP_SUMMARY tube to make the Study Pool (SP). A volume of 1500 µL of CHEAR Reference Plasma SP:SAMPLEPREP_SUMMARY was used for the CHEAR Pool (CP). An aliquot of 50 µL serum (including SP:SAMPLEPREP_SUMMARY experimental samples, SP, and CP) was transferred to a new, pre-labeled Lo-Bind SP:SAMPLEPREP_SUMMARY Eppendorf tube. There were total 200 experimental samples, 24 SP samples, and 24 SP:SAMPLEPREP_SUMMARY CP samples in this study. The samples were extracted by 400 µL methanol with SP:SAMPLEPREP_SUMMARY 500 ng/ml tryptophan-d5 as internal standard via a multiple tube vortex mixer SP:SAMPLEPREP_SUMMARY for 2 min at 5000 rpm. Protein precipitate was pelleted using a centrifuge SP:SAMPLEPREP_SUMMARY operating at room temperature for 4 min at 16,000 rcf. A volume of 350 µl of SP:SAMPLEPREP_SUMMARY the supernatant was transferred into a pre-labeled 2.0 ml Low-bind Eppendorf SP:SAMPLEPREP_SUMMARY tube, and then dried by a SpeedVac overnight. For immediate analysis, 100 µL of SP:SAMPLEPREP_SUMMARY water-methanol solution (95:5, v/v) was added to reconstitute the dried SP:SAMPLEPREP_SUMMARY extracts, and the samples were thoroughly mixed on multiple tube vortex mixer SP:SAMPLEPREP_SUMMARY for 10 min at 5000 rpm. Samples were centrifuged at 4°C for 4 min at 16,000 SP:SAMPLEPREP_SUMMARY rcf. The supernatant was transferred to pre-labeled autosampler vials for data SP:SAMPLEPREP_SUMMARY acquisition by LC-MS. Broad-spectrum metabolomics was conducted using a Thermo SP:SAMPLEPREP_SUMMARY Scientific™ Vanquish™ UPHPLC - Q Exactive™ HF-X Orbitrap System. SP:SAMPLEPREP_SUMMARY Metabolites were separated on an Acquity UPLC HSS T3 C18 (2.1 X 100 mm, 1.8 µm) SP:SAMPLEPREP_SUMMARY operating at 50 C using a reversed phase gradient separation with Water with SP:SAMPLEPREP_SUMMARY 0.1% Formic Acid (v/v) as mobile phase A and Methanol with 0.1% Formic Acid SP:SAMPLEPREP_SUMMARY (v/v) as mobile phase B. A 5 µL was injected into the instrument, and MS was SP:SAMPLEPREP_SUMMARY collected between 50-750 m/z in positive mode with the MS/MS data triggered by SP:SAMPLEPREP_SUMMARY the Data-Dependent Acquisition.
