Pregnant women were eligible if they were older than 18 ST:STUDY_SUMMARY years of age, English-speaking and had plans to deliver at the hospital in which ST:STUDY_SUMMARY they enrolled. Eligibility for ECHO-PATHWAYS included the following: recruitment ST:STUDY_SUMMARY at a Washington study site, consent to contact for future research, at least one ST:STUDY_SUMMARY pregnancy urine sample available, pregnancy questionnaire data collected and ST:STUDY_SUMMARY child age between 4–8 years old. GAPPS-PW has two study sites, Seattle, ST:STUDY_SUMMARY Washington, and Yakima, Washington. GAPPS is conducting two middle childhood ST:STUDY_SUMMARY visits – one at age 5-6 years and another at 8-9 years. The research is ST:STUDY_SUMMARY supported by NIH (UG3/UH3OD023271, P30ES007033, UL1 TR002319). The NC HHEAR Hub SP:SAMPLEPREP_SUMMARY thawed and transferred 50 µL of the study samples to a new set of tubes and SP:SAMPLEPREP_SUMMARY used them for the analysis. An additional 10 µL was taken from the original SP:SAMPLEPREP_SUMMARY study sample and transferred to another tube to make the total study pool for SP:SAMPLEPREP_SUMMARY this project, and then distributed with 50 µL per aliquot, used as quality SP:SAMPLEPREP_SUMMARY control study pools (QC study pools) throughout the whole analysis. All sample SP:SAMPLEPREP_SUMMARY aliquots (50 µL each) and QC study pools (50 µL each) were stored at -80° C SP:SAMPLEPREP_SUMMARY until the day of sample preparation. HHEAR Urine Pool 1 (50 µL each), HHEAR SP:SAMPLEPREP_SUMMARY Urine Pool 2 (50 µL each), CHEAR reference urine (50 µL each) and NIST urine SP:SAMPLEPREP_SUMMARY (SRM 3672) reference material (50 µL each) were provided by the NC HHEAR Hub. SP:SAMPLEPREP_SUMMARY LC-MS grade water (50 µL) was used as blanks. All samples were thawed at 4°C SP:SAMPLEPREP_SUMMARY overnight before the preparation. Samples, including study samples, study pool SP:SAMPLEPREP_SUMMARY samples, HHEAR reference urine, CHEAR reference urine, NIST reference urine, and SP:SAMPLEPREP_SUMMARY blanks were mixed with 400 µL methanol containing 500 ng/mL tryptophan-d5 as SP:SAMPLEPREP_SUMMARY internal standard and vortexed by a multiple tube vortex mixer for 2 min at 5000 SP:SAMPLEPREP_SUMMARY rpm at room temperature. All samples were centrifuged at 16,000 rcf for 10 min SP:SAMPLEPREP_SUMMARY at 4°C. The supernatant (350 µL) was transferred into a pre-labeled 2.0 mL SP:SAMPLEPREP_SUMMARY Lo-bind Eppendorf tube, dried by a SpeedVac overnight, and stored at -80° C. SP:SAMPLEPREP_SUMMARY For immediate analysis, 100 µL of water-methanol solution (95:5, v/v) was used SP:SAMPLEPREP_SUMMARY to reconstitute the dried extracts. Samples were thoroughly mixed on a multiple SP:SAMPLEPREP_SUMMARY tube vortex mixer for 10 min at 5000 rpm at room temperature and then SP:SAMPLEPREP_SUMMARY centrifuged at 4°C for 10 min at 16,000 rcf.
