Pregnant women receiving prenatal ST:STUDY_SUMMARY care at health centers in the Northeastern United States were recruited at 23.6 ST:STUDY_SUMMARY + 9.4 weeks’ gestation. Women were eligible if they were English or Spanish ST:STUDY_SUMMARY speaking, 18 years or older, did not have definite plans to move out of the area ST:STUDY_SUMMARY within a year after consent, were planning to give birth at the hospital ST:STUDY_SUMMARY affiliated with the recruitment site, had a singleton pregnancy, endorsed ST:STUDY_SUMMARY drinking <7 alcoholic drinks per week prior to pregnancy recognition and none ST:STUDY_SUMMARY following pregnancy recognition, had an HIV negative status, and delivered an ST:STUDY_SUMMARY infant without congenital abnormalities that would impede ongoing participation. ST:STUDY_SUMMARY Written informed consent was obtained from women in their preferred language. In ST:STUDY_SUMMARY 2016, funding was obtained to support ongoing prenatal recruitment and follow-up ST:STUDY_SUMMARY as part of the Environmental influences on Child Health Outcomes (ECHO) program ST:STUDY_SUMMARY through UH3OD023337. The NC HHEAR Hub thawed and transferred 50 µL SP:SAMPLEPREP_SUMMARY of the study samples to a new set of tubes and used them for the analysis. An SP:SAMPLEPREP_SUMMARY additional 10 µL was taken from the original study sample and transferred to SP:SAMPLEPREP_SUMMARY another tube to make the total study pool for this project, and then distributed SP:SAMPLEPREP_SUMMARY with 50 µL per aliquot, used as quality control study pools (QC study pools) SP:SAMPLEPREP_SUMMARY throughout the whole analysis. All sample aliquots (50 µL each) and QC study SP:SAMPLEPREP_SUMMARY pools (50 µL each) were stored at -80° C until the day of sample preparation. SP:SAMPLEPREP_SUMMARY HHEAR Urine Pool 1 (50 µL each), HHEAR Urine Pool 2 (50 µL each), CHEAR SP:SAMPLEPREP_SUMMARY reference urine (50 µL each) and NIST urine (SRM 3672) reference material (50 SP:SAMPLEPREP_SUMMARY µL each) were provided by the NC HHEAR Hub. LC-MS grade water (50 µL) was used SP:SAMPLEPREP_SUMMARY as blanks. All samples were thawed at 4°C overnight before the preparation. SP:SAMPLEPREP_SUMMARY Samples, including study samples, study pool samples, HHEAR reference urine, SP:SAMPLEPREP_SUMMARY CHEAR reference urine, NIST reference urine, and blanks were mixed with 400 µL SP:SAMPLEPREP_SUMMARY methanol containing 500 ng/mL tryptophan-d5 as internal standard and vortexed by SP:SAMPLEPREP_SUMMARY a multiple tube vortex mixer for 2 min at 5000 rpm at room temperature. All SP:SAMPLEPREP_SUMMARY samples were centrifuged at 16,000 rcf for 10 min at 4°C. The supernatant (350 SP:SAMPLEPREP_SUMMARY µL) was transferred into a pre-labeled 2.0 mL Lo-bind Eppendorf tube, dried by SP:SAMPLEPREP_SUMMARY a SpeedVac overnight, and stored at -80° C. For immediate analysis, 100 µL of SP:SAMPLEPREP_SUMMARY water-methanol solution (95:5, v/v) was used to reconstitute the dried extracts. SP:SAMPLEPREP_SUMMARY Samples were thoroughly mixed on a multiple tube vortex mixer for 10 min at 5000 SP:SAMPLEPREP_SUMMARY rpm at room temperature and then centrifuged at 4°C for 10 min at 16,000 rcf.
