CO:COLLECTION_SUMMARY            	heterospirocyclic compound, compound 25 (C25); (ii) the inactive precursor of
CO:COLLECTION_SUMMARY            	compound 25, PS027; (iii) atovaquone; and (iv) DMSO alone as the negative (no
CO:COLLECTION_SUMMARY            	drug) control. Following compound incubation (5 h), cultures were centrifuged at
CO:COLLECTION_SUMMARY            	2,000 RPM for 5 minutes, the media was removed and the cell pellets were washed
CO:COLLECTION_SUMMARY            	in ice cold PBS. All samples were centrifuged at 1,200g for 3 minutes, after
CO:COLLECTION_SUMMARY            	which metabolites were extracted from 5e7 cells using 100 µL of ice-cold
CO:COLLECTION_SUMMARY            	methanol extraction solvent. Samples were placed on a shaker at 4°C for 1 h and
CO:COLLECTION_SUMMARY            	spun at 13,000 RPM for 10 minutes at 4 °C. Supernatants were transferred into
CO:COLLECTION_SUMMARY            	high-performance liquid chromatography (HPLC) vials and stored at -80°C until
CO:COLLECTION_SUMMARY            	liquid chromatography-mass spectrometry (LC-MS) analysis. A pooled biological
CO:COLLECTION_SUMMARY            	quality control sample consisting of a 15 µL aliquot from each sample was also
CO:COLLECTION_SUMMARY            	generated for analytical quality control and to aid metabolite identification.

MS:MS_COMMENTS                   	Q-Exactive Orbitrap mass spectrometer (Thermo Scientific) coupled with
MS:MS_COMMENTS                   	high-performance liquid chromatography system (HPLC, Dionex Ultimate 3000 RS,
MS:MS_COMMENTS                   	Thermo Scientific) as described previously (Creek et al. 2016). Samples within
MS:MS_COMMENTS                   	the LC-MS batch were sorted according to blocks of replicates and randomized. To
MS:MS_COMMENTS                   	facilitate metabolite identification, approximately 350 authentic metabolite
MS:MS_COMMENTS                   	standards were analysed prior to the LC-MS batch and their peaks and retention
MS:MS_COMMENTS                   	time manually checked using the MZmine software. Pooled biological quality
MS:MS_COMMENTS                   	control samples and extraction solvent blanks were analysed periodically
MS:MS_COMMENTS                   	throughout the batch to monitor LC-MS signal reproducibility and aid downstream
MS:MS_COMMENTS                   	metabolite identification procedures. Raw LC-MS metabolomics data were analysed
MS:MS_COMMENTS                   	using the open source software, IDEOM
MS:MS_COMMENTS                   	(http://mzmatch.sourceforge.net/ideom.php). Briefly, the IDEOM workflow uses
MS:MS_COMMENTS                   	msconvert to convert raw files to mzXML format, XCMS (Centwave) to pick LC-MS
MS:MS_COMMENTS                   	peak signals and MZmatch for alignment and annotation of related metabolite
MS:MS_COMMENTS                   	peaks. Default IDEOM parameters were used to eliminate unwanted noise and
MS:MS_COMMENTS                   	artefact peaks. Confident metabolite identification was made by matching
MS:MS_COMMENTS                   	accurate masses to retention time of the ~350 authentic standards. When these
MS:MS_COMMENTS                   	authentic standards were unavailable, putative metabolite identification used
MS:MS_COMMENTS                   	accurate mass and predicted retention times. Metabolite abundance was
MS:MS_COMMENTS                   	represented by LC-MS peak height.
