The samples were kept cold and transported to the CO:COLLECTION_SUMMARY laboratory within the first two hours of the sampling. 5 mL of the breast milk CO:COLLECTION_SUMMARY samples were aliquoted in cryovials and stored at -80oC for glutamate analysis. CO:COLLECTION_SUMMARY Lactation stage 1 = colostrum, 2 = transition, 3 = mature. CO:SAMPLE_TYPE Milk #TREATMENT TR:TREATMENT_SUMMARY Mothers were recruited at Hospital Gineco Obstétrico Isidro Ayora in Quito, TR:TREATMENT_SUMMARY Ecuador. The participating mothers were permanent residents and indicated their TR:TREATMENT_SUMMARY willingness to exclusively breastfeed for at least two months. Mothers were TR:TREATMENT_SUMMARY healthy, ≥18 years of age, and primiparous, and they had not consumed TR:TREATMENT_SUMMARY antibiotics or probiotics during pregnancy. Each mother had given birth to a TR:TREATMENT_SUMMARY full-term baby via vaginal delivery. A total of 20 mothers donated their TR:TREATMENT_SUMMARY colostrum- (human milk from 1-7 days postpartum), transition- (15 days TR:TREATMENT_SUMMARY postpartum), and mature-milk (30 days postpartum). For Factor_2 (gender) in TR:TREATMENT_SUMMARY Study Design, 1: participant with a female baby; 2: one with a male baby; 0: not TR:TREATMENT_SUMMARY applicable. #SAMPLEPREP SP:SAMPLEPREP_SUMMARY Human breast milk samples were extracted using a two-phase extraction method. 2 SP:SAMPLEPREP_SUMMARY mL of chloroform-methanol (2:1) was added to each breast milk sample and SP:SAMPLEPREP_SUMMARY vortexed for 10 seconds. 600 µL of water was then added to each sample and the SP:SAMPLEPREP_SUMMARY mixture was centrifuged. 800 µL of the polar phase from each sample was SP:SAMPLEPREP_SUMMARY transferred into a 1.5mL glass tube. The samples were then dried using a speed SP:SAMPLEPREP_SUMMARY vacuum centrifuge. The polar phase was reconstituted in 700 µL of a phosphate SP:SAMPLEPREP_SUMMARY buffer (pH 7.4; 1M TSP). The samples were transferred to 5-mm NMR tubes for NMR SP:SAMPLEPREP_SUMMARY analysis.
