These resins contain diverse phenolic PR:PROJECT_SUMMARY metabolites, in particular hydroxycinnamate esters and methylated flavonoids. P. PR:PROJECT_SUMMARY trichocarpa leaf bud resin is characterized by methylated dihydrochalcone PR:PROJECT_SUMMARY aglycones. To determine how the resin profile is influenced by seasonal changes, PR:PROJECT_SUMMARY P. trichocarpa lateral leaf bud extracts (Total_resin_positive_mode.txt) and PR:PROJECT_SUMMARY secreted surface resin (Surface_resin_positive_mode.txt) were collected monthly PR:PROJECT_SUMMARY over a one-year cycle. The dihydrochalcones in both sets of extracts were PR:PROJECT_SUMMARY quantified using UPLC-MS and other chemical changes monitored using nontargeted PR:PROJECT_SUMMARY metabolomics by UPLC-HRMS. The results indicate that the dihydrochalcone content PR:PROJECT_SUMMARY changes over the seasons and that biosynthesis occurs concomitant with bud PR:PROJECT_SUMMARY development in the summer months. Non-targeted metabolomics data confirmed a PR:PROJECT_SUMMARY pattern of dramatic changes in the summer, and further suggested additional PR:PROJECT_SUMMARY periods of substantive biochemical change in the resin. While overall patterns PR:PROJECT_SUMMARY of surface-extracted resin matched that of whole bud extracts, some of the PR:PROJECT_SUMMARY dynamics were shifted in the surface resin samples. For leaf bud CO:COLLECTION_SUMMARY extraction, 10 lateral leaf buds were sampled from each biological replicate CO:COLLECTION_SUMMARY tree and frozen immediately in liquid nitrogen. For the collection of surface CO:COLLECTION_SUMMARY resin, three replicate sets of five similarly sized lateral leaf buds were CO:COLLECTION_SUMMARY collected from individual branches of each tree. Surface resin between May and CO:COLLECTION_SUMMARY July was collected from the entire leaf nodes, which included new buds as well CO:COLLECTION_SUMMARY as resinous expanding leaves, due to the very small size of the developing buds. CO:SAMPLE_TYPE Plant #TREATMENT TR:TREATMENT_SUMMARY The surface resin extracts of P. trichocarpa leaf buds were collected by TR:TREATMENT_SUMMARY accurately weighing 0.5 g (fresh weight) of intact lateral leaf buds and TR:TREATMENT_SUMMARY submerging them in 0.1 mL of HPLC-grade methanol per 10 mg of sample. The leaf TR:TREATMENT_SUMMARY buds were stirred for 1 min, after which the supernatant was collected. The TR:TREATMENT_SUMMARY methanol extract was filtered using a 0.20 μm PTFE filter to remove any solid TR:TREATMENT_SUMMARY particles. The extract was dried by transferring 2 mL of methanol extract into TR:TREATMENT_SUMMARY pre-weighed Eppendorf tubes and evaporating the samples using an Eppendorf TR:TREATMENT_SUMMARY concentrator until dry. For whole bud extracts, harvested leaf buds were TR:TREATMENT_SUMMARY homogenized into a fine powder in liquid nitrogen using a ceramic mortar and TR:TREATMENT_SUMMARY pestle and stored at -80°C before extraction. The powder was accurately weighed TR:TREATMENT_SUMMARY to 40 mg (fresh weight) and 1 mL of methanol was added. Samples were vortexed, TR:TREATMENT_SUMMARY sonicated for 10 min in a sonicating water bath, and centrifuged for 10 min at TR:TREATMENT_SUMMARY 15000 rpm. The supernatant was collected, and the extraction was repeated on the TR:TREATMENT_SUMMARY remaining plant tissue pellet. Supernatants from both extractions were combined, TR:TREATMENT_SUMMARY and the pooled samples were dried using a SpeedVac for 2 h until dry.
