CO:COLLECTION_SUMMARY            	Each sample represents metabolites extracted from 1E8 Plasmodium falciparum

CO:COLLECTION_SUMMARY            	Plus.

#TREATMENT
TR:TREATMENT_SUMMARY             	The II3 gna1-loxP line was tightly synchronized over a 5-hour window, using
TR:TREATMENT_SUMMARY             	sequential 70% Percoll centrifugation followed by sorbitol lysis, and
TR:TREATMENT_SUMMARY             	immediately treated with 10 nM rapamycin or DMSO (control) for one hour.
TR:TREATMENT_SUMMARY             	Rapamycin or DMSO were then removed from the culture medium and the parasites
TR:TREATMENT_SUMMARY             	were incubated until 57 hours post-Percoll when a new sorbitol synchronization
TR:TREATMENT_SUMMARY             	was performed at the beginning of cycle 1. The samples were then incubated for
TR:TREATMENT_SUMMARY             	about 81-89 hours post-Percoll and then enriched by MACS. For High Performance
TR:TREATMENT_SUMMARY             	Liquid Chromatography-Mass Spectrometry (HPLC-MS) analysis, 1x108 parasites per
TR:TREATMENT_SUMMARY             	replicate were replated in a 6-well plate and incubated for 2.5 hours to allow
TR:TREATMENT_SUMMARY             	recovery.
#SAMPLEPREP
SP:SAMPLEPREP_SUMMARY            	At the time of sample collection, after recovery from purification, the parasite
SP:SAMPLEPREP_SUMMARY            	metabolism was quenched through addition of ice-cold PBS. Parasites were
SP:SAMPLEPREP_SUMMARY            	pelleted (500 g, 4°C, 7 min) and metabolites were extracted from the pellet
SP:SAMPLEPREP_SUMMARY            	with 1 ml ice cold 90% methanol, vortexed 30 seconds, and centrifuged for 10 min
SP:SAMPLEPREP_SUMMARY            	at maximum speed (16000 x g) at 4°C. Samples were treated identically and
SP:SAMPLEPREP_SUMMARY            	swiftly to ensure reproducible results. The methanol supernatants were stored at
SP:SAMPLEPREP_SUMMARY            	-80°C until analysis, when they were transferred to fresh 1.5 mL tubes, dried
SP:SAMPLEPREP_SUMMARY            	down completely under nitrogen gas flow, and the metabolite residues stored at
SP:SAMPLEPREP_SUMMARY            	-70°C.
