Here we found that the transition of P. falciparum infection PR:PROJECT_SUMMARY from asymptomatic towards symptomatic malaria was associated with a >10-fold PR:PROJECT_SUMMARY reduction in the ratio of circulating unconjugated bilirubin over parasite PR:PROJECT_SUMMARY burden. Genetic deletion of Blvra in mice suppressed bilirubin production and PR:PROJECT_SUMMARY precipitated malaria mortality, owed to higher parasite burden and virulence. PR:PROJECT_SUMMARY Inhibition of bilirubin conjugation by the repression of hepatic UDP PR:PROJECT_SUMMARY glucuronosyltransferase family 1 member A1 (UGT1A1), increased the levels of PR:PROJECT_SUMMARY circulating unconjugated bilirubin and prevented malaria mortality. Unconjugated PR:PROJECT_SUMMARY bilirubin targeted P. falciparum directly inside red blood cells (RBC) to PR:PROJECT_SUMMARY suppress mitochondrion pyrimidine synthesis and inhibit the formation of PR:PROJECT_SUMMARY hemozoin crystals, compromising the parasite´s food vacuole´s capacity to PR:PROJECT_SUMMARY detoxify heme and extract essential amino acids (AA) from hemoglobin.
