The arachidonic acyl chain in PI is PR:PROJECT_SUMMARY introduced by lysophospholipid acyltransferase 11 (LPLAT11)/membrane-bound PR:PROJECT_SUMMARY O-acyltransferase 7 (MBOAT7), the loss of which causes microcephaly in humans PR:PROJECT_SUMMARY and mice. Here, we show that LPLAT11 deficiency impaired indirect neurogenesis PR:PROJECT_SUMMARY in the developing neocortex, resulting in fewer layer II-V neurons. PR:PROJECT_SUMMARY LPLAT11-deficient radial glial cells had defects in differentiation into PR:PROJECT_SUMMARY intermediate progenitor cells and increased apoptosis. Prior to these anomalies, PR:PROJECT_SUMMARY LPLAT11 deficiency caused a rounding of the Golgi apparatus, accompanied by PR:PROJECT_SUMMARY impaired apical trafficking of E-cadherin, and deregulated apical detachment. In Mboat7 KO ST:STUDY_SUMMARY mice, PI4P with arachidonic acid (38:4 PI4P), which is the major species in ST:STUDY_SUMMARY Mboat7 heterozygous mice, significantly decreased, and instead, PI4P with ST:STUDY_SUMMARY monounsaturated fatty acid (34:1, 34:2, 36:1, 36:2 PI) and PI4P with ST:STUDY_SUMMARY docosapentaenoic acid (DPA) or docosahexaenoic acid (DHA) (38:6, 40:5, 40:6, ST:STUDY_SUMMARY 40:7 PI4P) increased. PI(4,5)P2 also showed changes in the molecular species ST:STUDY_SUMMARY similar to those observed in PI4P. Interestingly, the total amount of PI(4,5)P2 ST:STUDY_SUMMARY started to decrease at E12.5 in Mboat7 KO mice, whereas the total amount of PI4P ST:STUDY_SUMMARY was unchanged. This is because the decrease of arachidonic acid-containing ST:STUDY_SUMMARY PI(4,5)P2 species was not fully compensated by the increase of other PI(4,5)P2 ST:STUDY_SUMMARY species. To rule out the possibility that changes in systemic lipid metabolism ST:STUDY_SUMMARY in global Mboat7 KO mice affect radial glial cells in a non-cell-autonomous ST:STUDY_SUMMARY manner, we generated telencephalon-specific Mboat7 KO mice (hereafter, ST:STUDY_SUMMARY Mboat7-Emx1-KO) by crossing Mboat7fl/fl mice with Emx1-Cre transgenic mice, in ST:STUDY_SUMMARY which Cre expression starts at E10.5 in the dorsal telencephalon. The number of ST:STUDY_SUMMARY cleaved caspase-3-positive cells increased in E13.5 and E14.5 cortices of ST:STUDY_SUMMARY Mboat7-Emx1-KO mice but not as much as in Mboat7 KO mice. Moreover, whereas ST:STUDY_SUMMARY p-H3-positive RGCs were located at the ventricular wall at E12.5, some ST:STUDY_SUMMARY p-H3-positive RGCs were located apart from the ventricular zone from E13.5 ST:STUDY_SUMMARY cortices of Mboat7-Emx1-KO mice (Fig 8C and D). Taken together, these results ST:STUDY_SUMMARY indicate that telencephalon-specific LPLAT11 deficiency caused the apical ST:STUDY_SUMMARY detachment and apoptosis of RGCs. In Mboat7-Emx1-KO mice, the change in the ST:STUDY_SUMMARY fatty acyl composition of PI and PIPs was delayed compared to Mboat7 KO mice, ST:STUDY_SUMMARY probably because Emx1-Cre drives the expression of Cre recombinase only from ST:STUDY_SUMMARY E10.5. Therefore, we generated neural-specific Mboat7 KO mice (hereafter, ST:STUDY_SUMMARY Mboat7-Sox1-KO) by crossing Mboat7fl/fl mice with Sox1-Cre transgenic mice, ST:STUDY_SUMMARY which drives the expression of Cre recombinase before E8.5 in the ST:STUDY_SUMMARY neuroepithelium. Detachment of RGCs from the ventricular wall was not observed ST:STUDY_SUMMARY in the E12.5 cortices of Mboat7-Sox1-KO. On the other hand, E13.5 cortices of ST:STUDY_SUMMARY Mboat7-Sox1-KO showed detachment from the ventricular wall and apoptosis to the ST:STUDY_SUMMARY same extent as those of Mboat7 KO mice. Also, Mboat7-Sox1-KO showed rounding of ST:STUDY_SUMMARY the Golgi apparatus at E12.5 when the detachment of RGCs from the ventricular ST:STUDY_SUMMARY wall was not observed, which is consistent with the phenotype observed in E12.0 ST:STUDY_SUMMARY cortices of Mboat7 KO mice. Moreover, E-cadherin overexpression failed to rescue ST:STUDY_SUMMARY the detachment of RGCs from the apical ventricular wall. In the Mboat7-Sox1-KO ST:STUDY_SUMMARY cortices, a significant change in the fatty acyl composition of PI and PIPs was ST:STUDY_SUMMARY observed. The total amount of PI(4,5)P2 started to decrease at E12.5 in the ST:STUDY_SUMMARY Mboat7-Sox1-KO cortices as well as in the Mboat7 KO cortices. The single-phase sample (a mixture of 170 µl of an aqueous SP:SAMPLEPREP_SUMMARY sample, 10 ng of internal standards [17:0/20:4 PI, 17:0/20:4 PI4P, and 17:0/20:4 SP:SAMPLEPREP_SUMMARY PI(4,5)P2], and 750 µl of quench mix) were mixed with 725 µl of CHCl3 and 170 SP:SAMPLEPREP_SUMMARY µl of 2 M HCl, followed by vortex-mixing and centrifugation (1,500 g, 5 min at SP:SAMPLEPREP_SUMMARY room temperature). The lower organic phase was collected into a new 2 ml SP:SAMPLEPREP_SUMMARY safe-lock poly-propylene tube and mixed with 708 µl of pre-derivatization wash SP:SAMPLEPREP_SUMMARY solution, followed by vortex-mixing and centrifugation (1,500 g, 3 min at room SP:SAMPLEPREP_SUMMARY temperature). The lower phase was collected into another fresh tube and SP:SAMPLEPREP_SUMMARY subjected to derivatization. Fifty µl trimethylsilyl diazomethane in hexane (2 SP:SAMPLEPREP_SUMMARY M solution) was added to the lipid extracts (~1 ml), and after leaving to stand SP:SAMPLEPREP_SUMMARY 10 min at room temperature, the reaction was quenched with 6 µl of acetic acid. SP:SAMPLEPREP_SUMMARY Next, 700 µl post-derivatization wash solution was added to the mixture, SP:SAMPLEPREP_SUMMARY followed by centrifugation (1,500 g, 3 min). The resultant lower phase was SP:SAMPLEPREP_SUMMARY collected and rewashed with a 700 µl post-derivatization wash solution. Samples SP:SAMPLEPREP_SUMMARY were temporarily stored at -80 ºC. Ninety µl of methanol and 10 µl of H2O SP:SAMPLEPREP_SUMMARY were added to the final collected lower phase. The samples were dried up with a SP:SAMPLEPREP_SUMMARY centrifugal evaporator. Samples were dissolved in 80 µl methanol, sonicated for SP:SAMPLEPREP_SUMMARY 1 min, and 20 µl H2O was added. After centrifugation at 15,000 g for 30 min at SP:SAMPLEPREP_SUMMARY 4 ºC, the supernatants were collected. To avoid degradation, the samples were SP:SAMPLEPREP_SUMMARY stored at -80 ºC until use. MultiQuant (SCIEX) was used for data MS:MS_COMMENTS quantification. Gaussian smoothing width was 2.0 points. Retention time of PI4P MS:MS_COMMENTS and PI(4,5)P2 is confirmed by measuring standards [17:0/20:4 PI3P, PI4P, PI5P, MS:MS_COMMENTS PI(3,4)P2, PI(3,5)P2, and PI(4,5)P2] before measuring samples. Peak areas of MS:MS_COMMENTS PI4P and PI(4,5)P2 were integrated from the ion chromatogram of PIP and of PIP2, MS:MS_COMMENTS respectively, based on retention time, and isotopic correction was applied. The MS:MS_COMMENTS peak area of the lipid species was divided by the corresponding internal MS:MS_COMMENTS standard area, and the total amount of phosphoinositides per cortex was MS:MS_COMMENTS calculated from the ratio of injection volume to total sample volume.
