Project Title:Untargeted Lipidomics of African American women at 8-10 months postpartum

Project Summary:Lipid dysregulation is hallmark of CMCPreg, specifically polyunsaturated fatty acids (PUFA) metabolism and downstream lipid mediators. To investigate whether this lipid dysregulation is prolonged after delivery, we performed untargeted lipidomics on serum collected 8-10 months postpartum from African American women who were diagnosed with cardiometabolic complications of pregnancy and from those with uncomplicated, term deliveries. We identified 11 lipids that were significantly increased in the postpartum serum of African American women who experienced cardiometabolic complications of pregnancy. These species belonged to sphingolipid, glycerolipid, and cholesteryl ester lipid classes and reveal dysregulated activity of glycerophospholipid metabolism, sphingolipid metabolism, sphingolipid signaling pathways, ferroptosis, and others by lipid enrichment pathway analysis. We also observed increases in Resolvin D3, 13 HpODE, AEA, OEA, and LEA suggesting dysregulated endocannabinoid signaling and increased phospholipaseA2 activity. Combined, these findings demonstrate that African American women who experienced cardiometabolic complications of pregnancy may have dysregulated lipid metabolism that extends as long as 8-10 months postpartum.
