Study Title | Analysis of DJ-1 Knockout Mouse Brains |
Study Type | Mouse genotype comparison |
Study Summary | We analyzed metabolites in the brains of wild type mice and DJ-1 knockout mice. The DJ-1 knockout mouse is a model of an inherited form of Parkinson's disease. |

Project Title: | Metabolic Response to the Loss of DJ-1 |
Project Type: | Comparison of the metabolic responses to the loss of DJ-1 in model organisms |
Project Summary: | Mutations in the DJ-1 gene cause rare forms of autosomal recessive Parkinson's disease in humans, and increased levels of DJ-1 protein are often found in cancers. Thus, understanding the precise function of DJ-1 is of broad interest. We hypothesized that analyzing differences in metabolites between organisms with and without DJ-1 may lead to clues about the protein's function. Here, we present the results of our ongoing studies using multiple different model organisms. |

Collection Summary: | -- |
Sample Type: | Brain - Cerebral Hemisphere |
Treatment:
Treatment ID: | TR000294 |
Treatment Summary: | The animals were either wild type of homozygous DJ-1 knockout animals. At 4 months of age, the mice were euthanized and their brains were processed for metabolomic analysis. |
Animal Vet Treatments: | None |
Animal Fasting: | No dietary interventions |
Animal Endp Euthanasia: | The mice were euthanized with CO2 followed by cervical dislocation. |
Animal Endp Tissue Coll List: | Cerebral Hemispheres |
Animal Endp Tissue Proc Method: | Immediately after euthanization, the brains were removed and dissected on an ice cold stage. The cerebellum was removed and the two cerebral hemispheres were bisected along the longtiduinal fissue. The cerebral hemispheres were immediately frozen on dry ice in individual tubes. The frozen tissue was then sent to Metabolon for subsequent processing. |
Sample Preparation:
Sampleprep ID: | SP000288 |
Sampleprep Summary: | Frozen cerebral hemispheres were shipped to Metabolon and processed there. A detailed summary of their methods is unavalible. |
