Collection Summary:Blood were aseptically collected from mice directly from the heart then left at room temperature for a few hours to clot, then centrifuged to separate the serum. Serum was then packed on dry ice and sent to the WCMC center in Davis, CA.

Treatment Summary:Mice were placed into 1 of 4 treatment groups, with three mice in each group. The first group contained uninjured mice who were not infected. The second group contained thermally injured mice who were not infected. The third group contained mice that were uninjured and infected with Pseudomonas aeruginosa. The fourth group of mice were thermally injured and infected with Pseudomonas aeruginosa.

Sample Preparation:

Sampleprep Summary:30µL of plasma was aliquoted into a 1.5 mL eppendorph tube. 1 mL of 3:3:2 ACN:H20:IPA was added to each sample. Samples were vortexed for 10 seconds and then shaken at 4°C for 5 minutes using an orbital mixing plate. Samples were centrifuged for 2 minutes at 14,000 rcf. 2 x 475 µL aliquots were removed, one dried down and submitted to derivitization, the other frozen at -20°C for backup. Samples were derivatized by adding 10 µL of MeOX and then shaken at 30°C for 1.5 hours using a orbital mixing plate. 91 µL of MSTFA + FAMES mixutre is added to each sample. Samples were shaken at 37°C for 30 minutes. Samples submitted to GC analysis.
