Sample Preparation:

Sampleprep Summary: | Half brains were homogenized with Dounce tissue homogenizers to make 10% (w/v) homogenate. The homogenate is then subject to ultracentrifuge at 100,000xg for 1h at 4°C, the resulting supernatant was neutralized with Tris buffer (0.5M, pH6.8) one tenth the volume of the supernatant. Then the neutralized supernatant is passed through 3kDa molecular weight cut-off filters at 4000 rpm for 45 mins at 4°C. Filtered supernatant was tried at SpeedVac Vacuum Concentrator at low heating until try pellets were received. Try pellets were then dissolved with 530ul of water and 60 ul of D2O containing 5mM of DSS. A final volume of 600ul for each sample was achieved. |
