Study SummaryTo investigate sex-dependent effects of fibroblast growth factor-21 (FGF21), diet-induced obese male and female wildtype mice were administered two acute doses of FGF21 or saline vehicle. Mice were euthanized, livers collected, and submitted for untargeted metabolomics analysis.

Treatment Summary:Male and female C57BL/6J mice were placed on 60% high-fat diet (Research Diets, Cat# D12492) for approx. 10 weeks beginning at 10 weeks of age. Mice were then counterbalanced into two treatment groups based on body weight (n = 9-11 mice/ sex/ treatment). For experiment, one dose of FGF21 (0.1 mg/kg, ip) or saline vehicle was given right before the onset of the dark cycle, then another dose (0.1 mg/kg, ip) was given the following morning after the onset of the light cycle. Mice were euthanized two hours following the AM injection, livers collected, and frozen in liquid N2.
