Study SummaryEx vivo hearts were pre-perfused with or without insulin, followed by ischemia induced by the cessation of perfusion. After 25 minutes of ischemia, hearts underwent 30 minutes of reperfusion. Right atrial tissue was then collected for IC-MS analysis. Three groups were included: a control group without ischemia, a group with 25 minutes of ischemia followed by reperfusion, and a group pre-perfused with insulin for 30 minutes prior to ischemia and reperfusion.

Project Title:Insulin mitigates acute ischemia induced atrial fibrillation and sinoatrial node dysfunction ex vivo
Project Summary:Acute atrial ischemia is a well-known cause of postoperative atrial fibrillation (POAF). However, the mechanisms through which ischemia contributes to the development of POAF are not well understood. In this study, ex vivo Langendorff perfusion was employed to induce acute atrial ischemia and reperfusion in the heart, resulting in a temporary form of POAF. The likelihood of atrial fibrillation (AF) was evaluated using programmed electrical stimulation and confirmed through open-atrium optical mapping. Compared to the control group without ischemia, 25 minutes of ischemia significantly increased the incidence of AF. The right atrium was found to be more susceptible to AF than the left atrium. Administering insulin for 30 minutes before ischemia and during reperfusion after 25 minutes of ischemia greatly reduced the vulnerability to AF. Additionally, IC-MS was utilized to assess the effect of insulin on the metabolism of the ischemic and reperfused heart.

Sampleprep Summary:Tissue were snap frozen in liquid nitrogen, then homogenized with Precellys Tissue Homogenizer. Certain volumes of homogenate were used to make sure same amount of tissue were extracted. Metabolites were extracted using ice-cold 80/20 (v/v) methanol/water with 0.1% ammonium hydroxide. Extracts were vortexed vigorously for 2 mins, centrifuged at 17,000 g for 10 min at 4°C, and supernatants were transferred to clean tubes, followed by evaporation to dryness under nitrogen. Dried extracts were reconstituted in 100 μL of 1mM potassium hydroxide (KOH) in deionized water.
