Catalytic ActivityATP + 1,2-diacyl-sn-glycerol = ADP + 1,2- diacyl-sn-glycerol 3-phosphate. {ECO:0000269|PubMed:12810723}.
FunctionPhosphorylates diacylglycerol (DAG) to generate phosphatidic acid (PA). Plays a key role in promoting cell growth. Activates the Ras/B-Raf/C-Raf/MEK/ERK signaling pathway induced by EGF. Regulates the recruitment of RAF1 and BRAF from cytoplasm to membranes and their heterodimerization. {ECO:0000269|PubMed:12810723, ECO:0000269|PubMed:19710016}.
PtmPhosphorylated; does not inhibit catalytic activity. {ECO:0000269|PubMed:12810723}.
Sequence CautionSequence=BAC05280.1; Type=Erroneous initiation; Note=Translation N-terminally extended.; Evidence={ECO:0000305};
SimilarityBelongs to the eukaryotic diacylglycerol kinase family. {ECO:0000305}.
SimilarityContains 1 DAGKc domain. {ECO:0000255|PROSITE- ProRule:PRU00783}.
SimilarityContains 1 PH domain. {ECO:0000255|PROSITE- ProRule:PRU00145}.
SimilarityContains 1 SAM (sterile alpha motif) domain. {ECO:0000255|PROSITE-ProRule:PRU00184}.
SimilarityContains 2 phorbol-ester/DAG-type zinc fingers. {ECO:0000255|PROSITE-ProRule:PRU00226}.
Subcellular LocationCytoplasm. Cell membrane. Note=Translocated from the cytoplasm to endosomes in response to stress stimuli. Isoform 2 is rapidly relocated back to the cytoplasm upon removal of stress stimuli, whereas isoform 1 exhibits sustained endosomal association. Translocates from the cytoplasm to the cell membrane in the presence of active GTP-bound form of HRAS.
SubunitIsoform 1 forms homooligomers through the SAM domain. Isoform 1 is also able to form heterooligomers with SAM domain- containing isoforms of DGKD. Oligomerization of isoform 1 inhibits its catalytic activity. Interacts with RAF1 and BRAF. {ECO:0000269|PubMed:12810723, ECO:0000269|PubMed:19710016}.
Tissue SpecificityIsoform 2 is ubiquitously expressed. Isoform 1 is expressed only in testis, kidney and colon. {ECO:0000269|PubMed:12810723}.
